Description
Bone marrow examination is useful in diagnosing and staging of hematologic disease, as well as assessing overall bone marrow cellularity. Due to ease of accessibility, aspiration, and biopsy, a bone marrow examination can also assist in the diagnosis of various storage and infiltrative disorders. ProPath’s Comprehensive Evaluation includes all the necessary testing to render a complete hematopathologic diagnosis on the patient’s blood and/or bone marrow and/or tissue specimen.

SPECIMEN REQUIREMENTS
Supplies for Submission
A ProPath-provided Bone Marrow Transport Kit contains the following:
  • Two 20-mL containers with 10% neutral buffered formalin
  • One EDTA (lavender topped) tube for molecular diagnostics
  • Two sodium heparin (green topped) tube for cytogenetics and flow cytometry
  • Two 5-slide capacity plastic slide holders
  • One large specimen bag
  • One small specimen bag
  • One requisition form
Specimen Collection
Refer to instructions in Collection of Bone Marrow Specimens.
Storage Requirements
Store specimens at room temperature (15 to 30°C) until transported. DO NOT freeze.
GENERAL AND TECHNICAL INFORMATION
Turn Around Time
Part 1 – Preliminary report with morphology and flow cytometry – 1-2 days
  • Morphology: 1 day (2 days if immunohistochemical staining is necessary)
  • Flow cytometry: 1-2 days
Part 2 – Final report with all remaining ancillary testing and conclusion – 5-10 days
  • Cytogenetics: 5-10 days (Longer time is usually required for karyotypes with complex clonal abnormalities)
  • FISH (Fluorescent in situ hybridization): 3-5 days
  • Molecular tests: 4-8 days
Methodology
  1. Morphology with/without cytochemistry and immunohistochemistry – hematoxylin and eosin stains for bone marrow biopsy and aspirate clot; Wright-Giemsa stain for aspirate, imprint, and blood
  2. Flow cytometry and Cytogenetics with/without FISH
  3. Molecular tests
Comprehensive Evaluation of Bone Marrow includes the evaluation of bone marrow morphology. If deemed necessary by the hematopathologist, flow cytometry, cytogenetics, and/or molecular diagnostics will be employed to render a concise, complete diagnosis.
Limitations
  • Please submit all specimens within 24 hours of collection for optimal results. Even if older than 24 hours, send them anyway; successful results have been obtained up to seven days after collection. Never discard a bone marrow specimen.
  • Send specimens Monday-Friday only.
